Organizing Your First Tissue Culture
Learn how to organize a source plant, create explants, record stages, and build a complete tissue culture history from day one.
Start with a Plant Record
Every tissue culture line traces back to a single source plant, and preserving that lineage is what turns later comparisons — which batch grew well, which line kept contaminating — into something you can actually learn from instead of a string of disconnected notes. Before taking any tissue, record the source plant's identity: a name, its species, and a photo if you have one. In xPlant this becomes a Plant record, the permanent parent for every explant and culture line taken from it, kept in your lab history even after every explant derived from it has been discarded. Create one from Dashboard → Plants → New Plant.
Tip: Use a consistent naming convention from the start, such as species + cultivar + batch or date. It makes finding and organizing plants much easier as your collection grows.
Establish an Explant
A plant record on its own doesn't grow in culture — the explant, a specific piece of tissue taken from that plant, does. Each explant needs its own identity because it develops its own independent culture history from the moment it enters media, even while staying linked back to its parent plant. What matters most here is a descriptive label — Shoot Tip 1, Node A — that will still make sense months later when comparing outcomes across a dozen explants from the same source. In xPlant, add one from the plant detail page via Add Explant.
Tip: Create one explant for each tissue source or culture you want to track independently. This gives every culture its own complete history.
Record Initiation
The first stage in an explant's life is initiation — the point where excised tissue is introduced to media and either establishes cleanly or shows early signs of contamination or stress. This is the moment most worth documenting carefully, since early observations often predict how a line performs later. Record the date, any observations, and the room or bench location if your lab tracks physical space. In xPlant, this becomes the explant's first Stage entry.
Tip: Record observations at every stage, even if they're brief. A note like "Healthy initiation, no browning" becomes valuable reference material later.
Record the Next Stage
Tissue culture progresses through discrete, named stages — Initiation, Multiplication, Rooting, Acclimatization — and each transition deserves its own record rather than an edit to the last one. Capture the date the culture entered the new stage, what you observed, and which media it's growing in, since media composition often explains why two otherwise identical explants diverge. In xPlant, this becomes a new Stage entry on the explant, so the full sequence stays visible, not just the current state. Add it from the explant detail page via Add Stage.
Tip: Attach a media recipe and SOP whenever possible. Together, they create a reproducible record of how the culture was grown.
Record a Transfer
Whenever culture material is split, moved to a new vessel, or combined with another line, that movement is a transfer — the thread that keeps a propagation history traceable instead of a series of disconnected stage updates. What's worth recording is where the material came from, where it went, and when. In xPlant, transfers are logged separately from stages so you can see exactly how one culture became several, or several became one, over time. Record one from the explant detail page or the dashboard's Transfers section.
Tip: Transfers are optional. If your lab only tracks stage progression, simply adding new stages may be all you need.
Review the Culture History
Once a few stages and transfers exist, the value of recording them shows up in the review: spotting which protocols are working, catching a line trending toward contamination, or comparing two explants from the same plant that took different paths. Every explant keeps its complete history — previous stages, observations, and transfers are never overwritten as a culture progresses. In xPlant, the dashboard surfaces this as a live view of active cultures, current stages, and any contamination alerts, with search and filters to find a plant or explant by name, species, stage, or label.
Tip: Review culture histories regularly to identify successful protocols, compare outcomes, and improve future tissue culture methods.
Recordkeeping principles
- Start simple: a complete record beats a perfect one. Begin with the plant, the explant, and its first stage, then layer in media, SOPs, and comments as the workflow becomes familiar.
- Record contamination as data, not a failure to hide. Log it as soon as you notice it, on the plant or explant it affected, so patterns across batches and media recipes become visible over time.
- Build records over time: a culture's value compounds the longer its history runs. A single stage entry tells you little; a full sequence from initiation through acclimatization tells you what actually worked.
- Keep recordkeeping consistent: the same labeling and observation habits across every explant are what make comparisons possible later. Pick a convention early and keep using it, even for cultures that seem too simple to bother.
